چکیده

C57BU10 (BlO; H2b ) mouse liver grafts are accepted in C3R (H2k) recipients without immunosuppressive tberapywhereas skin and heart grafts are rejected acutely.1 However, in vitro mixed leukocyte reactions (MLR) and the generation of cytotoxic T lymphocytes (crL) show the persistence of anti-donor responsiveness.2 The mechanism underlying this "split tolerance" phenomenon is unclear. The present study was undertaken to test the hypothesis that split tolerance might be due to the elimination of effector CfL through apoptosis, while CfL precursors, unsusceptible to apoptosis persist, as shown by the MLR and CfL responses in vitro.

Evidence for Deletion of Cytotoxic T Cells and Implications for Tolerance

MHC-mismatched liver grafts are accepted spontaneously between many mouse strains. The underlying mechanism(s) is un· clear. In the 810 (H2b ) to C3H (H2k) strain combination used in this study, donor T cells within the liver were rapidly replaced within 2 to 4 days of transplantation with those of the recipient. Freshly isolated liver graft.infiltrating cells harvested on days 4 and 7 exhibite...

Effect of Mesenchymal Stem Cells on ILT3 Expression in the Splenocytes of Skin Graft Recipient Mice

Background: Mesenchymal stem cells (MSCs) are considered as effective therapeutic cells in transplantation due to their immunomodulatory activities. However, precise mechanism of MSCs immunomodulatory activity is not completely understood. Objectives: To study the role of Immunoglobulin-like transcripts-3 (ILT3) immunomodulatory receptor in immune tolerance induced by MSCs in skin transpla...

بررسی تأثیر سرم موش حامله بر روی سلولهای دندریتیک در القاء تحریک لنفوسیت‌های T و تولید سیتوکین‌های IL-10 و IFN-γ Dendritic Cells and Antigen Specific T Cell Responses: Effect of Pregnant Mouse Serum

Background & Aim: Tolerance to the semi-allogenic fetal graft by the maternal immune system is a medical enigma that has stimulated investigations for a half of century. Several hypotheses have been proposed to explain the tolerance of mother to the fetus. The successful pregnancy is proposed and proved by many scientists to be a Th2 dominant phenomenon. This hypothesis is proved in most as...

Cardiomyocytes display low mitochondrial priming and are highly resistant toward cytotoxic T‐cell killing

Following heart transplantation, alloimmune responses can cause graft rejection by damaging donor vascular and parenchymal cells. However, it remains unclear whether cardiomyocytes are also directly killed by immune cells. Here, we used two-photon microscopy to investigate how graft-specific effector CD8(+) T cells interact with cardiomyocytes in a mouse heart transplantation model. Surprisingl...

Decreased expression of the interleukin 2 receptor on CD8 recipient lymphocytes in intestinal grafts rendered tolerant by liver transplantation in rats.

BACKGROUND In a previous study, it was shown that a spontaneously tolerated DA (RT1a) liver allograft in a PVG (RT1c) recipient was able to induce tolerance of a DA small bowel graft performed 17 days later in spite of infiltration of the intestinal grafts by mononuclear cells. AIMS To compare the phenotype of graft infiltrating cells in rejecting and tolerated small bowel grafts in order to ...
